Possible drug interactions
Some drugs/medications may interact with Tadalafil. Patients who have a condition called “left ventricular outlet obstruction” from valvular problems or heart muscle enlargement may get side effects such as fainting or light-headedness. Because patients with recent heart attacks or stroke, heart pains (angina), heart failure, uncontrolled blood pressure or uncontrolled irregular heart beats, severe liver disease, and retinal eye problems have not been studied, Tadalafil (Cialis) is not recommended for these patients. Researchers found that sildenafil, which was approved by the Food and Drug Administration (FDA) in 2013, decreased the levels of nitric oxide in the penile tissue. The drugs also reduced the production of cGMP. In the study, sildenafil reduced the cGMP levels by more than 60 percent, while tadalafil had the same effect. The drugs were effective in increasing penile rigidity in the sildenafil-treated rats. Sildenafil and tadalafil were taken orally, and the rats then ingested them with a mixture of saline and sildenafil.
What sets sildenafil apart from tadalafil is its ability to selectively inhibit phosphodiesterase type 5 (PDE5), a key enzyme in the erectile response. In the current study, sildenafil, tadalafil, and placebo were shown to inhibit PDE5, the enzyme responsible for cGMP degradation. Sildenafil, tadalafil, and placebo also improved erectile function in male rats. Sildenafil had the least side effect profile, such as decreased urinary flow or erections. The drugs also had little or no impact on vaginal bleeding in the sildenafil-treated rats. These results indicate that sildenafil and tadalafil may be useful in treating men with ED who are not responding to conventional erectile dysfunction drugs.
The drugs are currently available only through licensed generic pharmaceutical companies. They are also available on prescription only and are typically prescribed off-label to treat conditions that are not listed in the manufacturer's patient information leaflet.
